NEK7 inhibition attenuates Aβ<sub>42</sub>-induced cognitive impairment by regulating TLR4/NF-κB and the NLRP3 inflammasome in mice

NEK7 is a serine/threonine kinase that regulates cell mitosis and the activation of nucleotide-binding oligomerization domain-like (NOD-like) receptor thermal protein domain associated 3 (NLRP3) inflammasome, related to neuroinflammation neuronal damage. The purpose this study was explore role mechanism in cognitive impairment Alzheimer’s disease (AD). BV2 cells, microglia line, treated with Aβ42. expression measured reverse transcription–quantitative polymerase chain reaction western blotting. An apoptosis kit used determine apoptotic rate. APPswe/PS1dE9 (APP/PS1) transgenic mice were as an vivo AD model. experimental infected sh-NEK7 lentivirus downregulate NEK7. Finally, inhibition alleviated APP/PS1 mice. In conclusion, suppresses Aβ42-induced apoptosis, inflammation stress, improves performance may be potential target for treatment.
